If you send out a challenge how long does it take for it to be accepted. or if you want to be sure of a game by scheduling two teams in one slot will it affect the rest of the scheduling

It's accepted whenever the other owner accepts it. Unless it's an Open challenge, in which case the sim sets you up with whatever other team out there has issued an Open challenge, usually within 12 hours. Does the system even let you issue two challenges in the same slot?

Yes you can challenge more than one team in the same slot but when one accepts theirs the other will immediately disappear
