If I waive someone on the 40-man roster, does that immediately open up a spot on the 40? Or do I have to wait until he clears waivers (or doesn't) before the spot becomes open?

If you designate him with waivers, that opens up the slot immediately. If you just waive him, it does not.

Yup even if he clears waivers you have to then manually take him off the 40. Designate removes him from all rosters immediately and clears the 40 slot.
