This is my example. My opponent has a player with a speed rating of 50 and  I have a choice of 2 guys at that position (in this case it happens to be SF) one with a defensive rating of 66 and speed of 40 while the other has a defensive rating of 40 and a speed rating of 48. Which one should I play. Not only this scenario but in general is it more important to match speed or put the player with a higher defensive rating on the other player?

If your sole intention is to get the best defense out there, I would play the 66/40 DEF/SPD guy over the 40/48 guy.

I'd play the guy with higher defense against him, especially if you play M2M. Largely because the difference is larger between defense than speed when comparing the 2 guys. As far as your last question goes, I'd play a guy with 60 def and 50 speed over a guy with 60 speed and 50 defense any day.
