If I have an injured player in the ML's, can I promote a player to temporarily take his spot without having a demotion penalty when I send him back down? And if so, how do I do it correctly so that I don't get a penalty.

When you demote him back to the minors, just make sure you do both roster moves (removing from DL and demoting to the minors) at the same time.
