I have always set my players fatigue at fairly fresh. If I have players who are clearly better than the backups should I be setting them at getting tired or even at tired?

If there's a significant difference between the 1 and the 2 I will drop the started down to "getting tired." I've never gone lower than that in the settings because I've never had a situation where I felt the No. 2 guy wasn't at least better than my No. 1 playing at 70%.

you can also try to reduce the onset of fatigue by playing slowdown...and if the drop-off is considerable then it might be a good gameplanning decision as well.

Rednu is the 70% a made up number or is it listed somewhere what percentage each level is at?

What I've got written down is that the "performance penalty" for the progression of fatigue levels is "0, 0, 10, 30, 50." I honestly don't have a clue what the origin of that statement was, because it was written some time ago (sometime after the engine was changed...maybe around the time they tweaked the "kids expect to play" percentage from where ever it used to be down to 60?) I can try to pin it down more and look through dev chats around that time if they're still available in archive and see if I can find it, but it could also have been from a developer thread or a ticket exchange that I or someone else had and posted at some point on the issue. All I can say right now is that, at the time, the source had enough credibility in my eyes that I actually bothered to write it down. That may or may not mean it is actually accurate, but it is what I've always used when trying to decide things. Sorry I can't be more precise. 

ETA: Found the dev chat threads and couldn't find it, but I bet it came somewhere in the aftermath of the 2/3/11 chat since there were a lot of comments regarding fatigue and its effects that came out of that particular chat.
