Does the game penalize for using a starter as a reliver (as it states it does for using a position player out of position)?

No, pitchers can be used however you want. In fact, it's a pretty popular strategy to use low-inning starters in the bullpen.

Most of my relievers are high IP/G, they usually hold up better vs fatigue. They can also spot start in high leverage games.

2011 Doug Fister's partial season just threw 9.1ip shutout innings in 90 pitches. Sadly the bullpen lost it for him in the 11th

In-Game Fatigue:

It is my understanding that a starting pitcher's pitches per game before penalty is reduced by 25%, when he is used in relief. I read it on the Internet. (If he gets 100 as a starter, as a reliever, it's 75.)
